Carbohydrate chains in glycoprotein pharmaceuticals have important roles for the expression of their biological activities. Therefore, development of an assessment method for the carbohydrate chains is an important parameter for quality control of glycoprotein pharmaceuticals such as newly developed therapeutic antibodies. In this report, we applied capillary electrophoresis with laser-induced fluorescence detection to the analysis of carbohydrate chains after releasing with glycoamidase followed by derivatization with 3-aminobenzoic acid. We found that four major oligosaccharides present in antibody pharmaceuticals were successfully separated with good resolution. The present method showed good precision in both migration times and relative peak areas, and gave comparable accuracy with that using a derivatization method with 8-aminopyrene-1,3,6-trisulfonate.  相似文献

The introduction of a flavin chromophore on the myoglobin (Mb) surface and an effective electron-transfer (ET) reaction through the flavin were successfully achieved by utilizing the self-assembly of heterostranded coiled-coil peptides. We have prepared a semiartificial Mb, named Mb-1alphaK, in which an amphiphilic and cationic alpha-helix peptide is conjugated at the heme propionate (Heme-1alphaK). Heme-1alphaK has a covalently bound iron-protoporphyrin IX (heme) at the N terminus of a 1alphaK peptide sequence. This sequence was designed to form a heterostranded coiled-coil in the presence of a counterpart amphiphilic and anionic 1alphaE peptide sequence in a parallel orientation. Two peptides, Fla(1)-1alphaE and Fla(31)-1alphaE, both incorporating a 10-methylisoalloxazine moiety as an artificial flavin molecule, were also prepared (Fla=2-[7-(10-methyl)isoalloxazinyl]-2-oxoethyl). Heme-1alphaK was successfully inserted into apomyoglobin to give Mb-1alphaK. Mb-1alphaK recognized the flavin-modified peptides and a two-alpha-helix structure was formed. In addition, an efficient ET from reduced nicotinamide adenine dinucleotide to the heme center through the flavin unit was observed. The ET rate was faster in the presence of Fla(1)-1alphaE than in the presence of Fla(31)-1alphaE or the equivalent molecule that has no peptide chain. These results demonstrate that the introduction of a functional chromophore on the Mb surface can be achieved by using specific peptide-peptide interactions. Moreover, the dependence of the ET rate on the position of the flavin indicated that the distance between the heme active site and the flavin chromophore was regulated by the three-dimensional structure of the designed polypeptide.  相似文献

Acid treatment of the alkylated products of (Va, Vb, and VIII) of piperidinols IVa and IVb, and tetrahydropyridine VII with β-bromoethylbenzene, afforded 1,2,3,4,5,6-hexahydro-8-hydroxy-2,6-methano-6,1 1-dimethyl-3-phenethyl-3-benzazocine (la) in good yield. Piperidinols Va and Vb were also obtained from the reaction of N-(3-methyl-3-pentenyl)-β-phenethylamine (IIb) with methyl 3-(4-methoxyphenyl)-2,3-epoxypropionate.  相似文献

Stereoblock poly(lactic acid) consisting of D- and L-lactate stereosequences can be successfully synthesized by solid-state polycondensation of a 1:1 mixture of poly(L-lactic acid) and poly(D-lactic acid). In the first step, melt-polycondensation of L- and D-lactic acids is conducted to synthesize poly(L-lactic acid) and poly(D-lactic acid) with a medium-molecular-weight, respectively. In the next step, these poly(L-lactic acid) and poly(D-lactic acid) are melt-blended in 1:1 ratio to allow formation of their stereocomplex. In the last step, this melt-blend is subjected to solid-state polycondensation at temperature where the dehydrative condensation is allowed to promote chain extension in the amorphous phase with the stereocomplex crystals preserved. Finally, stereoblock poly(lactic acid) having high-molecular-weight is obtained. The stereoblock poly(lactic acid) synthesized by this way shows a higher melting temperature in consequence of the controlled block lengths and the resulting higher-molecular-weight. The product characterization as well as the optimization of the polymerization conditions is described. The end-functionalization of living polymers with bases (methanol, benzylamine, diethyl sodiomalonate, and sodium methoxide) and organosilicon compounds [X ? Si(CH3)3;X ? : CH2?C(CH3)COO? , CH3COO? , CH2?CHCH2? , C6H5? ] was investigated in the living cationic polymerization of styrene initiated with the 1-phenylethyl chloride/SnCl4/nBu4NCl system in CH2Cl2 at ?15°C. The four bases and C6H5SiMe3, independent of their structures, were apparently incapable of reacting with the living end and invariably led to polystyrenes with the ω-end chlorine [~ ~ ~ CH2CH(Ph)Cl] originated from the initiating system. The number-average end-functionality (F?n) of the chloride, determined by 1H-NMR, was close to unity (F?n > 0.9). The presence of chlorine in the polymer was also confirmed by elemental analysis. In contrast, the quenching by the trimethylsilyl compounds with X = methacryloxy, acetoxy, and allyl gave ω-end-functionalized polystyrenes with the corresponding terminal groups (X) for which the F?n values were close to unity (F?n > 0.9). The chemical modification of proteins is an effective technique for manipulating the properties and functions of proteins, and for creating protein-based materials. The N-terminus is a promising target for single-site modification that provides modified proteins with uniform structures and properties. In this paper, a copper(II)-mediated aldol reaction with 2-pyridinecarboxaldehyde (2-PC) derivatives is proposed as an operationally simple method to selectively modify the N-terminus of peptides and proteins at room temperature and physiological pH. The copper(II) ion activates the N-terminal amino acids by complexation with an imine of the N-terminal amino acid and 2-PCs, realizing the selective formation of the nucleophilic intermediate at the N-terminus. This results in a stable carbon-carbon bond between the 2-PCs and the α-carbon of various N-terminal amino acids. The reaction is applied to four different proteins, including biopharmaceuticals such as filgrastim and trastuzumab. The modified trastuzumab retains the human epidermal growth factor receptor 2 recognition activity.  相似文献

Abstract

The polymerizations of alkyl esters of propiolic acid by Rh complex catalysts were investigated. [Rh(norbornadiene)Cl]2, which was the most active among the catalysts examined, gave rise to poly(alkyl propiolate) in a fairly high yield (~80%) in the presence of alcohol as the polymerization solvent. The polymers formed were a pale yellow powder soluble in common organic solvents except for poly(methyl propiolate). The structures of the polymers obtained were investigated by IR, 13C-NMR, CP MAS 13C-NMR, and laser Raman spectroscopies, together with the x-ray diffraction method. Based on these spectroscopic data, it was concluded that this Rh complex can be called a stereoregular polymerization catalyst of alkyl propiolate because the poly(alkyl propiolate) obtained has a cis-transoidal structure.  相似文献

Polymerizations of cyclooctene, 5-methyl, 5-chloro-, and 5-methoxycyclooctenes were studied. Cyclooctene (CO) and 5-methylcyclooctene (MCO) provided high polymers in 80% yield with the use of WCl6/AlEti.B Clu5 or WCl6/AlEtCl2 catalyst. 5-Chlorocyclooctene gave oligomer in 50% yield with WCl6/AlEt2Cl catalyst. Neither polymer nor oligomer was produced from 5-methoxycyclooctene. These polymers were found to be produced through a ring-opening mechanism. The ratio of cis to trans structure in poly(CO) and poly(MCO) was determined by measurements of the decoupled ′H-NMR spectrum. Poly(CO) containing more than 50% trans structure was a crystalline solid at room temperature, while the polymer containing 30% of trans structure did not crystallize at room temperature. Poly(MCO) was amorphous, regardless of the content of trans structure. Poly(CO) and poly(MCO) obtained with MoCU/AlEtaCl or MoCU/AlEtCb catalyst contained no carbon-carbon double bond, and a vinyl polymerization mechanism was expected for this system.  相似文献
